چکیده

Mononuclear manganese(II) and iron(III) flavonolates were synthesized as synthetic enzyme-substrate complexes, and their oxygenation reactions as biomimetic functional models with relevance to flavonol 2,4-dioxygenases are briefly described.

Probing the molecular determinants of aniline dioxygenase substrate specificity by saturation mutagenesis.

Aniline dioxygenase is a multicomponent Rieske nonheme-iron dioxygenase enzyme isolated from Acinetobacter sp. strain YAA. Saturation mutagenesis of the substrate-binding pocket residues, which were identified using a homology model of the alpha subunit of the terminal dioxygenase (AtdA3), was used to probe the molecular determinants of AtdA substrate specificity. The V205A mutation widened the...

Ring-cleaving dioxygenases with a cupin fold.

Ring-cleaving dioxygenases catalyze key reactions in the aerobic microbial degradation of aromatic compounds. Many pathways converge to catecholic intermediates, which are subject to ortho or meta cleavage by intradiol or extradiol dioxygenases, respectively. However, a number of degradation pathways proceed via noncatecholic hydroxy-substituted aromatic carboxylic acids like gentisate, salicyl...

Structure of the 2,4′-dihydroxyacetophenone dioxygenase from Alcaligenes sp. 4HAP

The enzyme 2,4'-dihydroxyacetophenone dioxygenase (DAD) catalyses the conversion of 2,4'-dihydroxyacetophenone to 4-hydroxybenzoic acid and formic acid with the incorporation of molecular oxygen. Whilst the vast majority of dioxygenases cleave within the aromatic ring of the substrate, DAD is very unusual in that it is involved in C-C bond cleavage in a substituent of the aromatic ring. There i...

Assessment Effect of Different Amounts of Iron and Manganese Sulfates on the Qualitative and Quantitative Yield of Wheat in South West of Iran (Ramhormoz Region)

In order to evaluate the effect of different amounts of iron and manganese sulfates on wheat yield (Chamran cultivar), a factorial experiment based a randomized complete block design with three replications. The experiment treatments consisted of different concentrations of iron sulfate )Including; zero, 1500, and 3000 grams per hectare) and manganese sulfate (Including; zero, 1500, and 3000 gr...

Intrinsic tryptophan fluorescence as a probe of metal and alpha-ketoglutarate binding to TfdA, a mononuclear non-heme iron dioxygenase.

2,4-Dichlorophenoxyacetic acid (2,4-D)/alpha-ketoglutarate (alphaKG) dioxygenase, TfdA, couples the oxidative decarboxylation of alphaKG to the oxidation of the herbicide 2,4-D using a mononuclear non-heme Fe(II) active site. The intrinsic tryptophan fluorescence associated with the four Trp residues in TfdA allows for the use of fluorescence spectroscopy to monitor the binding of iron and alph...
